Can someone tell me why I'm getting this error only when I run my script on startup and how to fix it?
The why is easy: the audio subsystem isn't started until a user logs in.
You've not said which of the many possible ways of running your python script you are using, nor have you said which OS you are using.
The fix is to ensure the audio subsystem is started before your python script gets run. Precise instructions depend on information you have not provided.
One approach would be to switch to using PulseAudio and run that as a System-Wide Daemon
